چکیده

At present, there are no compounds in clinical development in the field of chronic myeloid leukemia (CML) or Philadelphia-positive (Ph) acute lymphoblastic leukemia (ALL) that have been documented to harbor significant activity against the imatinib-resistant T315I mutation. Recent reports on the preclinical activity of some emerging tyrosine kinase inhibitors such as ON012380, VX-680 and PHA-739358 promise possible clinical efficacy against this specific Bcr-Abl mutant form. Here, we focus on the role of aurora kinase inhibitor VX-680 and PHA-739358 in blocking the leukemogenic pathways driven by wildtype and T315I-Bcr-Abl in CML or Ph ALL by reviewing recent research evidence. We also discuss the possibility of employing aurora kinase inhibitors as a promising new therapeutic approach in the treatment of CML and Ph ALL patients resistant to first and second generation TK inhibitors.

Analysis of Expression Of SIRT1 Gene In Patients With Chronic Myeloid Leukemia Resistant To Imatinib Mesylate

Background: Chronic myeloid leukemia is a clonal myeloproliferative disease which is characterized by bcr/abl translocation. With the emergence of tyrosine kinase inhibitors such as imatinib mesylate, significant improvement has been made in treatment of this disease. However, drug resistance against this medicine is still an obstacle. SIRT1 is a gene with deacetylase activity which has been de...

Pregnancy Outcome of Two Patients with Chronic Myelogenous Leukemia Treated with Imatinib

Although chronic myelogenous leukemia in pregnancy is rare, its management and treatment is more difficult and complicated.Treatment of patients with chronic myelogenous leukemia includes bone marrow transplantation, however in less than 30% of patients the donor’s organ would be accepted. To this end, cytotoxic therapy is considered as an alternative therapeutic option. This option provides sa...

Study of STAT3 Expression in Different Phases of Patients with Chronic Myeloid Leukemia

Background and Aim: Chronic myeloid leukemia(CML) is a clonal myeloproliferative disease, characterized by BCR/ABL translocation. Using tyrosine kinase inhibitors such as Imatinib, treatment for this disease has progressed remarkably. However, resistance to tyrosine kinase inhibitor is a major obstacle. Signal transducer and activator of transcription 3(STAT3) is an important transcription fact...

Sequential ABL kinase inhibitor therapy selects for compound drug-resistant BCR-ABL mutations with altered oncogenic potency.

Molecularly targeted kinase inhibitor cancer therapies are currently administered sequentially rather than simultaneously. We addressed the potential long-term impact of this strategy in patients with chronic myelogenous leukemia (CML), which is driven by the fusion oncogene BCR-ABL. Analysis of BCR-ABL genotypes in CML patients who relapsed after sequential treatment with the ABL inhibitors im...

Overcoming CML acquired resistance by specific inhibition of Aurora A kinase in the KCL-22 cell model.

Serine/threonine kinase Aurora A is essential for regulating mammalian cell division and is overexpressed in many types of human cancer. However, the role of Aurora A in chemoresistance of chronic myelogenous leukemia (CML) is not well understood. Using the KCL-22 cell culture model we have recently developed for studying mechanisms of CML acquired resistance, we found that Aurora A expression ...
